Finally, got to the root of the issue!!
As suggested i did use a multimeter, and found that there was no power getting to the LEDs. The cause of this was the barrel jack adapter that is being used to break out the barrel jack into wire terminals. There was no continuity between the negative side of this connection, between the outer ring and the negative terminal. This was resolved by cutting the barrel jack off the PSU and wiring all the wires directly via wago connectors.
Now the LED strip is receiving 5v and as soon as the Pi was powered back on every worked immediately and exactly as intended. Perfect!
